Buddleja officinalis의 추출물로 지각과민증 치료에 관한 연구

초록

Dentin hypersensitivity (DH) is defined as short and sharp pain caused by external stimuli such as heat, vaporization, contact, osmotic pressure, and chemical stimulation in a normal tooth, rather than due to disease or tooth damage. Its solution is to block the flow of dentinal fluid by physically blocking the dentinal tubule. Of these treatments, fluoride and oxalate type for hypersensitivity can only have a temporary effect. Resins should be used with a suitable bonding system and they may cause hypersensitivity symptoms after treatment. Overcoming these limitations, there is a need for method that can effectively treat dental hypersensitivity lasting long without any side effects. For this reason, experiments with 200 plant extracts as candidates for dentin hypersensitivity, Buddleja officinalis was considered as a candidate for present study. The purpose of this study is to confirm whether the ethanol extract of Buddleja officinalis is effective to protect enamel and dentin by coating tooth surface and resistance to acid or alkali even after tooth coating. uPAR 및 uPA의 RNAi 형질감염이 타액선 종양세포주에 미치는 영향

초록

Despite existing chemotherapy and surgical resection strategies, salivary gland adenocarcinoma(AdCa NOS) is one of the major causes of mortality among malignant salivary gland tumors. New therapeutic measure are needed to improve the outcome for patients with AdCa. Overexpression of urokinase-type plasminogen activator receptor/urokinase-type plasminogen activator(uPAR-uPA) has been implicated in progression and metastasis of oral cancer. RNA interference(RNAi) which has emerged as an effective method to target specific genes for silencing has provided new opportunities for cancer therapy. But there has been rarely reported using RNAi-uPAR/uPA transfection in salivary gland AdCa. The purpose of this study were to examine the specific inhibition of uPAR/uPA mRNA and protein expression by RNAi transfection of uPAR/uPA through RT-PCR and Immunoslot blot, and to study tumor cell proliferation activity, adhesion, invasion and migration of SGT cell line in vitro compared to the controls. In adhesion assay, cells transfected with RNAi-uPAR/uPA inhibited markedly adhesion to vitronectin compared to parental cells. Angiogenic assays revealed a significant decrease in the angiogenic potential of SGT cells downregulated by both uPAR and uPA. In migration assay, suppressing uPAR and uPA inhibited the capacity of the cells to migrate compared to parental cells. In invasion assay, cells transfected with RNAi-uPAR/uPA showed the maximum decrease in invasion when compared to all other treatment conditions. RNAi expressing plasmids efficiently downregulated mRNA and protein expression of uPAR and uPA. Cell cycle analysis showed that the simultaneous downregulation of uPAR and uPA caused the accumulation of cells in the sub-G0/G1 phase in SGT cells. Immunoslot blot analysis revealed that downregulation of uPAR and uPA caused the prominent activation of caspase 8. It suggested that the RNAi targeting of the uPAR/uPA system could have a therapeutic potentiality for malignant salivary gland tumors. 여드름의 원인 및 관리실태 분석

The need for skin health care is newly recognized in accordance with the importance and needs of skin care due to harmonious interpersonal relation. In this study, collected 202 questionnaire sheets in Seoul were analyzed by SPSS 18.0 program through data coding and data cleaning, and frequency analysis for general characteristics was done. Heredity, environmental factors and management condition of acne, and the differences according to general characteristics were analyzed by Crosstabs. Mainly, women used skin care shop and their age groups were varied from twenties to fifties. Recognition of skin trouble were compound(42.1%), and significant statistical difference(p<.05), while oily or compound to man and compound or acne skin to woman were observed. First occurrence of acne was generally middle school(33.7%). Facial acne progress areas was cheek(36.1%), forehead(30.7%), chin(28.2%) and neck(5.0%) in descending order, and present state of acne was in progress(60.9%). Etiologic factors were stress(39.1%), pre and post-menstruation(22.8%), environment(20.8%), heredity(17.3%). There was significant statistical difference between gender(p<.01). Treatment places were at dermatologist(45.0%) and special skin care shop(27.2%). The most reliable place for treatment was special skin care shop(44.6%) and dermatologist(42.6%). It suggested that in acne of skin trouble, age groups of customers could be varied among male and female and in order to meet such requirements of customers, high quality service ought to be provided so that acne care could satisfy customers with the differentiated and careful service.

허혈성 암세포에서 상피-간엽 이행과 연관된 유전자 발현 프로필

초록

Hypoxia is one of the most common features of cancer. It is also associated with cancer progression and the acquisition of aggressiveness, which includes invasion and metastasis. Oral squamous cell carcinoma accounts for 90% of all oral cancers, and its 5-year survival rate is about 50%. Despite various attempts and trials, its prognosis has not improved. Among numerous adverse prognostic factors, hypoxia is suspected as one of the most important factors, as it increases the aggressiveness of oral cancer cells. We attempted to observe the effect of hypoxia on the expression of epithelial-mesenchymal transition markers in oral cancer cells. We analyzed and compared both the mRNA and protein expression levels of epithelial-mesenchymal markers using qRT-PCR and western blotting in both normoxic and hypoxic YD10B oral squamous cell carcinoma cells. Eighty-six genes were analyzed through real-time PCR using commercial microarray plates, performed in triplicate. Among the 86 genes, the expression of 24 were increased (≥ 2 fold) by hypoxia, while that of three genes was decreased (≥ 2 fold). Hypoxia significantly affects epithelial-mesenchymal transition-related genes. Further studies on the regulation of these genes may help to develop more efficient therapeutic modalities for oral cancer and to improve prognosis of oral cancer patients. 비스포스포네이트와 연관된 악골괴사 부골에서의 세균 검출

The purpose of this study was to identify the oral bacterial species in sequestra from patients with bisphosphonate-related osteonecrosis of the jaw (BRONJ). Fifteen patients with BRONJ (2 males and 13 females) were evaluated. Clinical features, radiographic findings, and bisphosphonate intake history were investigated. All patients were treated with surgical methods (curettage or sequestrectomy). Infected bone samples were collected from the affected BRONJ site. Ten bacterial species were selected for polymerase chain reaction (PCR) detection. Two to nine bacterial species were detected by PCR. Gram-negative species were predominant and all identified bacteria were anaerobes. Porphyromonas gingivalis, Tannerella forsythia, and Treponema denticola were detected at high levels. These are major pathogenic species in periodontal disease. Orthopantomographic radiographs showed generalized alveolar bone loss in most patients. These radiographic findings may provide evidence of chronic periodontitis as a pre-existing inflammatory disease. Most patients had experienced a predisposing dental procedure, such as tooth extraction. Sequestra (necrotic bone) infected with oral bacterial species may be an important risk factor for BRONJ. As such, prevention and management of BRONJ may rely on effective control of bacteria in the oral cavity. Key words : BRONJ , Bacteria, Sequestra, Necrotic bone

RAW 264.7 세포에서 과산화수소에 의한 세포반응을 IP-HPLC로 확인하는 예비연구

Hydrogen peroxide (H2O2) is originally an endogenous small molecule which is reduced into water in cells. In order to know the H2O2-induced oxidative stress in RAW 264.7 cells, first of all, the optimum concentration of exogenous H2O2 which show reactive cellular responses was determined as 40 μM by MTT assay, and followed by 40 μM H2O2 application in RAW 264.7 cells for 30 min, 1, or 2 hours. The expressional changes of essential proteins for cellular proliferation, epigenetic modification, inflammation, apoptosis, survival, and protection were assessed by immunoprecipitation high performance liquid chromatography (IP-HPLC) using 51 antisera. 40 μM H2O2 treatment down-regulated proliferation-related proteins, Ki-67, PCNA, CDK4, cyclin D2, cMyc, and PLK4, induced histone methylation/ deacetylation and DNA methylation by increasing levels of HDAC10 and DMAP1 and by decreasing levels of DNMT1 and KDM4D, activated inflammatory reaction by increasing levels of MCP-1, COX-2, CD68, LTA4H, CXCR4, and lysozyme, and dramatically up-regulated cellular apoptosis-, survival-, and protection-related proteins, AIF, PARP-1, caspase 9, c-caspase 9, pAKT1/2/3, SOD-1, HO-1, NF-kB, NRF2, and GSTO1 in RAW 264.7 cells. These observations suggest exogenous 40 μM H2O2-induced oxidative stresses which resulted global cellular responses including not only antioxidant, inflammation, and apoptosis but also proliferation and epigenetic modification. Particularly, 40 μM H2O2-induced apoptosis was mainly derived from PARP-1/AIF signaling leading parthanatos, and 40 μM H2O2-induced suppression of cMyc/MAX/MAD network was relevant to reduction of RAW 264.7 cell proliferation. Accordingly, H2O2 appears to affect RAW 264.7 macrophages in several ways eliciting not only oxidative stresses but also genome-wide DNA damage. 상악에 재발한 거대한 치성 점액섬유종

초록

The myxoma(odontogenic myxoma, myxofibroma) of the jaws is a benign, usually slow-growing infiltrative tumor of connectice tissue. This article describes a case history of the treatment of recurred odontogenic myxofibroma in the maxilla and further discusses the appropriate management of such cases with reference to the literature review. A 55-year-old male patient presented with a gingival swelling in the right side of maxilla. He had history of second time recurred disease. Microscopically, a high cellularity fibrous tumor with fibroblastic cells was observed. Tumor cells were composed of stellate and spindle shaped fibroblasts. Islands of odontogenic epithelium were seen in the specimen. The final diagnosis was made with odontogenic myxofibroma in consideration of twice-time recurrences and histological characteristics. Because of the high likelihood of recurrence, an accurate differential diagnosis of pathology at the time of initial visit is very important in determining treatment options, and immunohistochemical tests may be helpful. 악안면 부위에 발생한 혈관종 및 혈관기형에 대한 후향적 연구

The vascular origin diseases occurring in the oral and maxillofacial region can be divided into hemangiomas and vascular malformation. They are rare and early lesions are not easy to diagnosis, so they may be misdiagnosed as clinicians, or the method and timing of treatment may be inappropriate. For 10 years, from August 2005 to August 2015, we analyzed the clinical records, diagnosis, treatment, and prognosis of 23 patients with hemangioma or vascular malformation confirmed by clinical, histopathological or radiological examination of maxillofacial lesions at department of oral and maxillofacial surgery, Pusan National University Hospital and Pusan National University Dental Hospital. Chief complants of patients were mainly swelling, mass, and spot-like lesion on various intraoral sites. Treatments were performed with excision, embolization, sclerotherapy, depending on each lesions condition. Arteriovenous malformation cases were more frequently recurred than other lesions. Prognosis of lesions were relatively good but close observation should be needed. Key words : Maxillofacial area, Benign tumor, Vascular malformation, Hemangioma

Ninjurin 1이 골모세포의 분화에 미치는 영향

Nerve injury induced protein 1 (Ninjurin1) was originally described in neuroscience in which the expression of Ninjurin1 was regulated by Schwann cells and dorsal root ganglion neuronal cells of damaged nerve tissues. After the first discovery of Ninjurin1, the widespread expression of Ninjurin1 in adult and embryonic tissues have been observed including bone marrow, peripheral blood lymphocytes, thymus, and heart. Currently, the Ninjurin1 mediated positive regulation of pre-osteoclasts fusion and osteoclast development was reported. The bone homeostasis is dynamically balanced by bone-resorbing activity of osteoclast and bone-forming activity of osteoblast. Until now, the role of Ninjurin1 was never been described in osteoblastogenesis. Therefore, in this study, we have evaluated the expression and function of Ninjurin1 in osteoblast. The ample expression of Ninjurin1 was observed in bone marrow of mouse tibia sections but it was barely expressed in osteocytes. And also the expression levels of Ninjurin1 were gradually increased during osteoblast differentiation of calvarial pre-osteoblast, C2C12, and MC3T3-E1 cells. Importantly, the expression of Ninjurin1 was increased in the absence of osteogenic stimulus, BMP2, which suggests the cell density-dependent regulation of Ninjurin1. The controlled expression of Ninjurin1 by cell-density was evidently shown in not only pre-osteogenic osteoblast lineage cells but also in non-osteogenic cancer cells such as HeLa and A549 cells. In addition, the isoform-specific knockdown of Ninjurin1 remarkably reduced the alkaline phosphatase (ALP)-positive osteoblast differentiation. Thus, our results suggest a previously unappreciated mechanism of Ninjurin1 expression and also suggest its role on osteoblastogenesis. 당뇨환자와 비당뇨환자 간의 두경부 근막간극 감염 시 임상 및 병리학적 특징 비교

This study includes the clinical features of diabetic patients with fascial space infections of head and neck and those of non-diabetic patients. We retrospectively studied 62 patients who were treated for fascial space infections of the head and neck, and compared the patients’ background, the aetiology of the infection, clinical variables, laboratory values, and treatment process. Statistical analyses of the differences between groups were made with Student's t test, the chi square test, and analysis of variance, as appropriate. Diabetic patients had more spaces involved, had longer hospital stays, and developed more complications. The main microorganism of diabetic group was Klebsiella pneumonia and otherwise, Streptococcus spp. was for non-diabetic group. We conclude that diabetic patients are more likely to develop complications and it is necessary to consider about dominant microorganism when using medication for diabetes patients Key words : Facial space infection, Disbetes mellitus, Klebsiella pneumonia
